چکیده

Effects of repeated postnatal administration of caffeine (10 and 20mg/kg s.c. daily from P7 to P11) were studied in two models of epileptic seizures characterized by spike-and-wave EEG rhythm in 18- and 25-day-old rats. Rhythmic metrazol activity (RMA, model of human absences) was induced by low dose of pentylenetetrazol (PTZ, 20mg/kg or 40mg/kg, i.p.) and minimal clonic seizures (model of human myoclonic seizures) by two successive doses of PTZ (20 and 40mg/kg i.p.). Early postnatal caffeine treatment resulted in significant changes of RMA only in 18-day-old rats. Anticonvulsant effects were observed in RMA episodes elicited by the 20-mg/kg dose of PTZ in both caffeine groups whereas latency of RMA episodes induced by the 40-mg/kg dose was shortened and their duration was prolonged. No changes were found in 25-day-old animals. Incidence, EEG and motor pattern of minimal clonic seizures were not changed. Some animals in both control age groups exhibited transition to generalized tonic-clonic seizures. This type of seizures never appeared in caffeine-treated 25-day-old animals. Mixed effects of postnatal caffeine exposure were demonstrated; these predominantly anticonvulsant effects are age- and model-specific.

Different effects of postnatal caffeine treatment on two pentylenetetrazole-induced seizure models persist into adulthood.

BACKGROUND Postnatal treatment with caffeine from P7 to P11 (10 or 20 mg/kg daily) resulted in transient changes in two pentylenetetrazole (PTZ)-induced models of epileptic seizures characterized by spike-and-wave EEG rhythm in immature rats. To know if some changes persist into adulthood we studied these models in young adult Wistar rats. METHODS Caffeine treatment at a daily dose of 10 and/...

مقایسه اثر تجویز مزمن مورفین و ترامادول در دوره شیرخواری بر تشنج حاد حاصل از پنتیلن تترازول در موش صحرایی نابالغ

Background: It is demonstrated that morphine and tramadol affects seizure but the mode of action of these drugs on seizure has not been compared yet with increasing of age. The aim of this study was to compare the impact of exposure to these drugs on Pentylenetetrazol-induced seizure in immature rat. Methods: Male neonate rats were randomly chosen and divided into three groups namely Saline (n=2...

اثر تزریق مزمن مرفین در دوره نوزادی بر تشنج حاصل از پنتیلن تترازول در دوره قبل از بلوغ موش‌های صحرایی

Background & Aims: Frequent administration of morphine leads to increase in motor activity, motivational properties, and other responses related to this kind of drugs.  It has been reported that the impact of morphine on seizure is dose dependent. The aim of this study was to study the impact of chronic exposure to morphine on pentylenetetrazol-induced seizure in rats. Materials & Methods: Neon...

Altered expression of orexin 1 and endocannabinoid 1 receptors of the hippocampus in three pentylenetetrazol, pilocarpine and kainate seizure models

Introduction: Seizure is synchronous and abnormal brain neuronal activity that leads to activation of different receptors capable of enhancing or suppressing seizure activity such as orexin receptor 1 (OXR1) and/or endocannabinoid receptor 1(CBR1). The time of activation for the receptors may influence seizure control. Therefore, this study aimed to investigate the latency for and the change of...

The effects of pentylenetetrazol, chlordiazepoxide and caffeine in rats tested in the elevated plus-maze depend on the experimental illumination.

The so-called anxiolytic and anxiogenic drugs are considered to cause, respectively, increases and decreases in plus-maze open arm exploration, without modifying locomotor activity occurring in the closed arms in an elevated plus-maze when the animals are tested in an illuminated environment. Simply testing animals in the dark also increases open arm exploration, which may be interpreted as an ...
